FACED v3.0 Ji Lab
Designed and developed a more stable and wide-range covering excitation wavelength 2P integrated free-space angular chirp enhanced delay (FACED) microscope v3.0 for high-speed volumetric neural imaging. This project was launched to overcome the inherent design limitations of the prior system: significant thermal and mechanical instability from its off-the-shelf parts, and a fixed 1035nm laser system that was incompatible with SOTA genetically encoded sensors optimized for other wavelengths.
- System Architecture & Design: Designed the complete system architecture from the ground up to maximize stability. Modeled all optical paths using ray-tracing simulations in Zemax OpticStudio and designed all custom, high-stability mechanical mounts in Autodesk Inventor, using more stable materials for mounts to reduce thermal and mechanical drift (everything was precision machined and designed for fixed mounting). Managed all component sourcing, vendor relations, and procurement.
- Construction & Integration: Led a multi-disciplinary team through the physical construction and subsystem integration, culminating in a fully commissioned instrument that met all initial design specifications.
- Data Acquisition Framework: Oversaw the electronic control for the data acquisition framework. Supervised the adaptation and validation of existing LabVIEW software for seamless integration with a new laser system, and verified all hardware interfaces to ensure robust, phase-locked operation.
- System Alignment & Characterization: Directed the final system alignment and characterization, empirically validating key performance metrics against design simulations to confirm near diffraction-limited resolution and sub-micron mechanical stability.
- Data Analysis Pipeline: Developed in-house data analysis pipelines for the post-processing of image stacks from FACED to extract biological data from the high-throughput, multi-terabyte datasets generated by the microscope, enabling quantitative analysis of experimental results.
